RESUMO

Respiratory syncytial virus (RSV) is an enveloped non-segmented negative sense RNA virus that belongs to Orthopneumovirus genus of the Pneumoviridae family in the order Mononegavirales. The virus is the leading cause of severe respiratory disease in children under two years of age and is responsible for substantial disease burden in infants and elder people in both developed and developing countries1,2. RSV is only known to circulate among humans, though it was first isolated from chimpanzees3. The virus can experimentally infect mice, rats, cotton rats, ferrets, and hamsters, but does not naturally circulate in these animal populations4. We found that Malayan pangolins (Manis javanica) were naturally infected with RSVs that have 99.4-99.8% genomic identity with strains circulating in humans. Phylogenetic analyses revealed that five RSVs in pangolins were RSV-A ON1 and seven were RSV-B BA genotypes, both of which are currently prevalent in humans worldwide. These findings suggest that humans might transmit their viruses to endangered wildlife.

PURPOSE: To study the changes of microarchitecture of alveolar bone due to different duration of ovariectomy in rats. METHODS: Twenty-four virgin Sprague-Dawley rats were randomly assigned to ovariectomy group (OVX) or sham-ovariectomy group (sham). OVX rats were subjected to bilateral ovariectomy and sham-ovariectomy was conducted in sham rats. Six rats of each group were sacrificed respectively 3 months and 6 months after surgery. The right semi-maxilla of all rats were scanned by Micro-CT, and the inter-radicular alveolar bone of the maxillary first molar was analyzed. Statistical analysis was carried out with SPSS 16.0 software package. RESULTS: Two-dimensional and three-dimensional reconstructed images of the alveolar bone showed porotic changes in rats both 3 months and 6 months after ovariectomy, including thinner, looser trabeculae and expanded bone marrow. When compared with corresponding sham rats, BMD, BV/TV and Tb.Th of alveolar bone significantly decreased in OVX rats both 3 months and 6 months after ovariectomy (P<0.05). Tb.N and Tb.Sp significantly increased in both OVX groups (P<0.05). When compared with 3 months after ovariectomy, the rats 6 months after ovaroectomy shared deceased BMD, BV/TV and Tb.Th (P<0.05) and increased Tb.N (P<0.05). CONCLUSIONS: Bone loss and deterioration of trabeculae of alveolar bone aggravates with the extended duration of ovariectomy in OVX rats.

PURPOSE: The osteogenic-angiogenic differentiation effects of simvastatin (Sim) were explored on adipose tissue-derived stem cells (ASCs). A tissue-engineered bone with simvastatin loaded ß-tricalcium phosphate (ß-TCP) scaffold and ASCs was constructed to repair the calvarial defect in rabbits. METHODS: ASCs were obtained from the groin of rabbits. After 14 days of osteogenic inducing culture, sufficient cells were expanded for the following experiments. Cell counting was conducted to ASCs in osteogenic inducing medium containing 0, 0.01, 0.1 and 1 µmol/L simvastatin. Concentrations of 0.05 and 0.1 µmol/L simvastatin were administrated to ASCs for real-time PCR of angiogenesis-osteogenesis related genes like RUNX2, OPN, OCN, and VEGF on day 1, 7. ALP staining was performed on day 7, Alizarin red staining for calcium deposits was carried out on day 14. Bilateral critical-sized defects were created on 12 New Zealand rabbits. Four groups of tissue-engineered bone were randomly allocated to them. Group A: ß-tricalcium phosphate (ß-TCP) (n=6); group B: ß-TCP/Cell (n=6); group C: ß-TCP/Sim (n=6); group D: ß-TCP/Cell/Sim (n=6). Specimens were decalcified and stained by HE 8 weeks after operation. The data was statistically analyzed using SPSS 17.0 software package. RESULTS: The use of simvastatin with the concentration of 0.05 µmol/L enhanced the expression of angiogenic-osteogenic related genes like RUNX2, OPN, OCN, and VEGF. ALP activity and von Kossa were significantly stronger in osteogenic inducing medium containing 0.05 µmol/L simvastatin. The new bone formation area of ß-TCP/Cell/Sim group at 8-week after implantation was significantly larger than the other groups. CONCLUSIONS: 0.05 µmol/L simvastatin enhances the angiogenic-osteogenic differentiation of ASCs. Simvastatin loaded ß-TCP scaffold and ASCs successfully repair the calvarial defect in rabbits. These results indicate a promising future in application of simvastatin for bone regeneration.

An approach was proposed to evaluate preparation technology by means of fingerprint-peak matching technology of high Performance liquid chromatography with diode array detector (HPLC-DAD). Similarity and hierarchical clustering analysis (HCA) were applied to identify the 15 batches of Xiaochaihu granules from different manufacturers and our laboratory, and peak pattern matching between the composite formulae and Radix Bupleuri Chinensis, which was one of the main ingredients of Xiaochaihu granules, was utilized to evaluate the preparation technology of Xiaochaihu granules via the indexes of the relative deviation of retention time (RT) and UV spectrum feature similarity of their corresponding peaks. Eleven matching peaks were found between Xiaochaihu granules and Radix Bupleuri Chinensis. However, the saikosaponin A and saikosaponin D, which are the important active components in Radix Bupleuri Chinensis, were not found in Xiaochaihu granules from any manufacturers. The peak areas of 11 characteristic peaks of Xiaochaihu granules samples formed a matrix of 11 × 15. The result of HCA showed that Xiaochaihu granules samples were divided into four kinds of category. Xiaochaihu granules samples from the same manufacturer were basically clustered of the same category. The results suggested that the saikosaponin A and saikosaponin D are prone to structural transformation under the condition of decoction and in the presence of the organic acidic components. These active components, existing in raw herb, might transform to a series of non-active secondary saikosaponin due to unfavourable preparation technology. So the conventional decoction-based preparation technology of Xiaochaihu granules might greatly affect its quality and therapeutic effectiveness. This study demonstrates that fingerprint-peak matching technology can not only be used for quality control of this composite formulae, but also provide some guidance for preparation technology of Xiaochaihu granules.

OBJECTIVE: To study the inhibitory effect of erythritol by contrast to xylitol on growth and acid production of Streptococcus mutans (S. mutans). METHODS: S. mutans were incubated respectively in 0.5%, 1%, 2%, 4%, 8%, 12%, 16% erythritol or xylitol culture medium under anaerobic conditions. The A and pH value of the mediums were measured at 0, 2, 4, 6, 8, 10, 12, 18, 24 hours, following the profile plots by SPSS. RESULTS: The data of A were higher in 0.5%, 1%, 2% erythritol culture medium than in xylitol culture medium at the same concentration, while lower in 8%, 12%, 16% erythritol culture medium than in xylitol culture medium at the same concentration. The data of pH were lower in 0.5%, 1%, 2% erythritol culture medium than in xylitol culture medium at the same concentration, while higer in 8%, 12%, 16% erythritol culture medium than in xylitol culture medium at the same concentration. It indicated that the growth and acid production of S. mutans were higer in 0.5%, 1%, 2% erythritol culture medium than in xylitol culture medium at the same concentration, while lower in 8%, 12%, 16% erythritol culture medium than in xylitol culture medium at the same concentration. CONCLUSION: Compared with xylitol, erythritol in low concentration has weaker effort on the growth and acid production of S. mutans, while having stronger effort in high concentration.

It has been reported that there is an interaction between Benzo[a]pyrene (BaP), a widespread carcinogenic polycyclic aromatic hydrocarbon, and tributyltin (TBT), an organometal used as an antifouling biocide. This study was therefore designed to examine the potential in vivo influence of BaP, TBT and their mixture on splenic antioxidant defense systems of Sebastiscus marmoratus. The fish were exposed to water containing environmentally relevant concentrations of BaP, TBT and their mixture. Spleens were collected for biochemical analysis after exposure for 7, 25, 50 d and after recovery for 7, 20 d. Cotreatment with BaP and TBT for 7 d potentiated the induction of glutathione peroxidase (GPx) activity by BaP or TBT alone. The cotreatment for 25 and 50 d resulted in inhibition of GPx activity, which was similar to the effect of TBT. Splenic glutathione S-transferase (GST) activities were significantly elevated in S. marmoratus exposed to BaP starting from 7 d and remained high up to 25 d. However, no further activity change was found with prolonged exposure. Cotreatment of BaP and TBT primarily inhibited the GST activity, which was similar to the effect of TBT. Cotreatment with BaP and TBT for 25 or 50 d potentiated the depletion of GSH (glutathione) by BaP or TBT alone. MDA (malondialdehyde) contents in spleen of S. marmoratus were not significantly altered compared with the control during the test period. Spleen, as an immune organ, is sensitive to exposure of BaP or TBT. It should have an effective mechanism to counteract oxidative damage. Antioxidative defense systems in spleen of S. marmoratus should be considered as potential biomarkers. Short-term exposure of BaP or TBT could result in induction of antioxidant defense system. A significant decrease of these indices, such as GSH, GST, GPx might indicate more severe contamination.

RESUMO

A novel method for the preconcentration with crosslinked chitosan (CCTS) and determination of trace manganese in waters by flame atomic absorption spectrometry has been presented. The absorption rate of manganese (VII) by CCTS was 98% at pH 3.00. The effect of preconcentration time, dosage of CCTS, temperature, sample volume and co-existing elements have been investigated. The mechanism of adsorption of CCTS for Mn(VII) was also discussed. The detection limit of the method was 1.86 microg x L(-1), and the relative standard deviation (RSD) was 2.0% (n = 10). The method has been applied to the determination of trace manganese in south lake and Changjiang water with satisfactory results.
